Is java a good language?
i find java not so attractive. reducing everything to classes, interfaces %26amp; etc are very uneligant. compared to c, perl, vb, java sucks.

I just can't extract any DNA out of my B.subtilis culture? Why?
Refering to Cheng, H.R., and Jiang, N., (2006) "Extremely rapid extraction of DNA from bacteria and yeast", Biotechnology Letters, 28: 55-59.
I followed most of the protocol, except that I used P:C:I (25:24:1) instead of tris-saturated phenol.
1. 1ml of culture, centrifuged, washed with STE buffer twice.
2. Resuspend cell pellets with TE buffer
3. Add 1x volume of P:C:I (25:24:1), vortex for 60secs.
4. Centrifuge at max speed for 5mins at 4C.
5. Transfer aqueous layer to new tube, mix with 1x volume of chloroform, centrifuge at max speed for 5mins at 4C (repeat until no white interface is present)
6. Transfer aqueous layer to new tube, add RNase A, incubate at 37C for 30mins
7. Add 1x volume of chloroform, mix and centrifuge at max speed for 5mins.
8. Transfer aqueous layer (which theoretically should contain pure DNA) to new tube and store at -20C
Before RNAse A treatment, I can see a %26gt;10kb band n trace of RNA, however after the treatment, no band was seen. Why?
I just can't extract any DNA out of my B.subtilis culture? Why?
Either i) your cells are not lysing, ii) you are losing genomic DNA at step 3 (I am not a fan of doing the phenol and choloroform (PCI) at the same time, you save very little time doing this), iii) you are losing the DNA in step 5 (you need to make sure you transfer all of the aqueous layer and I often end up taking a very small amount of the white bottom layer also into the next step), or iv) your RNase is contaminated with DNase. I would try the extraction without the RNase step and see what you get. Also, are you digesting the DNA before gel electrophoresis (you would see a smear), and are you doing an ethanol precipitation at the end and reconsituting the DNA in buffer? If you are adding DNA from the chloroform extraction step directly to the agarose gel, you may be losing significant amounts at that step also.

Reply:Are you looking for RNA or DNA? At the end, you state that the *RNA* band was gone after RNAse treatment - which is not too surprising ;-)
If, however, you mean that your DNA weas gone after RNAse treatment: are you sure your RNAse isn't contaminated with any DNAse?
Also - the DNA is insoluble in the chloroform, so that final purification stage is just to remove the RNAse. Will you be needing RNAse-free DNA subsequently? If not, then you could try missing that stage out.
Only things I can think of off the top of my head.

since i am using computer from 5 to 6 years.i have good command over computers internet etc. now i want to start to learn programming from basic to advance. i dont have any knowledge of language fundamentals. i have seen VB 2005 i have designed some basic interfaces in it. but later i came to know that i must have good concept of OOPS or C or c++. few told me you cant learn VB2005 without learning vb 6 or Vb .net 2003. if i will start learning that it will take 6 or more months and than VB2005. its not look great for me. i dont want develop hardcore giant and multinetwork applications. i am planning to develop midrange of applications relating to inventory accounts etc. my question is

I need to create the following, which is pretty simple and I can create the HTML form, but I have no idea how to write the Python CGI script... (below part2)

Accessing mobile Gmail via IE?
I will be traveling to Central America this summer and last time I was there, I found (obviously) the internet connections extremely slow, and Gmail was almost prohibitively slow, taking sometimes 10 minutes to load so I could read my email. I had the idea of using the mobile Gmail interface (http://m.gmail.com) but I am unable to open this page in Internet Explorer, only Firefox. Since most computers I've been on in C.A. only have IE, I was wondering what's up, or if this problem is reproduceable for any of you out there. Thanks!

I pay about 90 cents per week for a good Internet Security Suite and I have no adware, spyware, malware, virus, trojan or whatever they throw at me.
In context I spend $30 a week on lattes, but I could just drink water.
Priorities sunshine.

Reply:WOW thats alot of bad stuff. i have a few cleanup and program suggestions. choose whichever you like. your computer safety programs might not be the safest depending on what they are. but i can guarentee my suggestions are safe. cleaning up your computer can make it run faster. also go to the start menu, then go to RUN, then type msconfig and choose the startup and you can enable or disable certain programs from starting when you boot your computer. but here are my suggestions:
*AVG is not as reliable as AVAST go to http://download.com and download it (search for Avast home) dont register it and when the demo is up, just uninstall it and reinstall it. it also helps because if you download a bad file from limewire or the internet, it alerts you immediately so you can delete it right away. certain AVG programs will be charging for updates soon and thats not good if you cant pay for them you ushould always keep your antivirus up to date. keep Avast home edition open in your toolbar at all times. it moniters alot of potentially harmful things its really great. if you dont have firewall i suggest downloading Zonealarm (which is free) and if you need antispyware, download Spybot search and destroy and superantispyware free edition
you might also want to download ccleaner to keep your computer cleaner make sure you keep any programs you download (that i suggested) updated
as for cleanup tips i
suggest the following; here are a few things to clean up your computer:first go to the start panel and click RUN then type %temp% then delete EVERYTHING in that file but not the file itself.(this will help you clean up your computer a little bit)
Also go to Mycomputer, Click drive C, then click the Windows Folder, then find the Prefetch Folder Open that (dont delete the folder itself) and delete all the items in that.

I pay about 90 cents per week for a good Internet Security Suite and I have no adware, spyware, malware, virus, trojan or whatever they throw at me.
In context I spend $30 a week on lattes, but I could just drink water.
Priorities sunshine.
A full Internet Security Suite is so cheap and just like insurance.
Reply:WOW thats alot of bad stuff. i have a few cleanup and program suggestions. choose whichever you like. your computer safety programs might not be the safest depending on what they are. but i can guarentee my suggestions are safe. cleaning up your computer can make it run faster. also go to the start menu, then go to RUN, then type msconfig and choose the startup and you can enable or disable certain programs from starting when you boot your computer. but here are my suggestions:
*AVG is not as reliable as AVAST go to http://download.com and download it (search for Avast home) dont register it and when the demo is up, just uninstall it and reinstall it. it also helps because if you download a bad file from limewire or the internet, it alerts you immediately so you can delete it right away. certain AVG programs will be charging for updates soon and thats not good if you cant pay for them you ushould always keep your antivirus up to date. keep Avast home edition open in your toolbar at all times. it moniters alot of potentially harmful things its really great. if you dont have firewall i suggest downloading Zonealarm (which is free) and if you need antispyware, download Spybot search and destroy and superantispyware free edition
you might also want to download ccleaner to keep your computer cleaner make sure you keep any programs you download (that i suggested) updated
as for cleanup tips i
suggest the following; here are a few things to clean up your computer:first go to the start panel and click RUN then type %temp% then delete EVERYTHING in that file but not the file itself.(this will help you clean up your computer a little bit)
Also go to Mycomputer, Click drive C, then click the Windows Folder, then find the Prefetch Folder Open that (dont delete the folder itself) and delete all the items in that.

** RAM **
Maximum RAM (amount and speed) is 1 GB of PC2700 (333-MHz) DDR SDRAM using 2 x 512 MB DIMMS. This isn't bad for XP. As long as you're upgrading, make certain you get the fast RAM and the max amount. Top-rated newegg RAM http://www.newegg.com/Product/ProductLis... is $58 shipped (CAS 2.5), but you may be able to get a better deal elsewhere. Also, check with Dell and make certain standard DDR SDRAM DIMMs (DIMM is the form factor) will fit in your motherboard - this seems to be true, judging by the specs.
** HARD DRIVE **
You have only IDE connectors, so you have to get an IDE hard drive. From the pictures, it *looks* like you can only have 1 hard drive, but you *may* be able to add a second where the floppies and optical drive go. You have 2 IDE channels, which means that you can attach 4 IDE drives total. That doesn't mean that there is enough room in the case for all 4! Judging by the specs, you have 4 drive bays total (2 optical, I'm guessing 1 floppy, and 1 hard drive). That means 1 floppy, the new DVD drive and both hard drives OR (if you want) 1 hard drive and 3 external drives (of whatever sort). Since you're upgrading the hard drive, there will probably be plenty of space on the new drive, so I would say keep your 3 externally accessible drives and (eventually) ditch the old hard drive and the CD-ROM (keep the CD-RW).

It's not important that you cannot run at full speed - all that's important is that these drives will work on your system. You may have ATA-33 or ATA-66. I *believe* that faster drives will work (at the slower speed).

Look at the "ultra ATA" listings at the left (if DELL has approved ATA-100 and ATA-133 for your system). Our 40 GB drive is available for $35 shipped, a Maxtor 120 GB 7200 is available for $40 shipped, and the 160 GB drive for $45 shipped. Clearly, you can keep going up - any of these drives should fit in your system, and you may want a 320 GB drive (though I don't know why). 250 GB looks like the most GB for the $ (Hitachi, $49 shipped, ATA-133, and 8 MB cache, which should make it even faster than the others). Avoid those "Magnetic Data" drives - this company is unknown to me. Make certain to get a 7200 RPM, and (again) make certain that ATA-100 and ATA-133 will work in your system.
** VIDEO **
Another problem is your video, which (I believe) uses shared RAM. Updating your video card should improve your game performance. You are restricted to a PCI bus (do *not* get anything that says PCIe or x16 or AGP). Again, pricegrabber is good for an overview

DVD writers (burners) are so cheap, there's no point in restricting yourself to a DVD-ROM. Again, E-IDE is the standard, so you will have to make certain that this will work with your IDE connectors.

However, since you didn't mention the price you'd like to pay, I believe you can go to http://support.dell.com/support/topics/g... and then enter your service tag. After that, you can find all the accessaries you want and you can go ahead to buy everything from DELL. Even if you don't like their prices, you at least have something to refer to and compare with.

** RAM **
Maximum RAM (amount and speed) is 1 GB of PC2700 (333-MHz) DDR SDRAM using 2 x 512 MB DIMMS. This isn't bad for XP. As long as you're upgrading, make certain you get the fast RAM and the max amount. Top-rated newegg RAM http://www.newegg.com/Product/ProductLis... is $58 shipped (CAS 2.5), but you may be able to get a better deal elsewhere. Also, check with Dell and make certain standard DDR SDRAM DIMMs (DIMM is the form factor) will fit in your motherboard - this seems to be true, judging by the specs.
** HARD DRIVE **
You have only IDE connectors, so you have to get an IDE hard drive. From the pictures, it *looks* like you can only have 1 hard drive, but you *may* be able to add a second where the floppies and optical drive go. You have 2 IDE channels, which means that you can attach 4 IDE drives total. That doesn't mean that there is enough room in the case for all 4! Judging by the specs, you have 4 drive bays total (2 optical, I'm guessing 1 floppy, and 1 hard drive). That means 1 floppy, the new DVD drive and both hard drives OR (if you want) 1 hard drive and 3 external drives (of whatever sort). Since you're upgrading the hard drive, there will probably be plenty of space on the new drive, so I would say keep your 3 externally accessible drives and (eventually) ditch the old hard drive and the CD-ROM (keep the CD-RW).
Pricegrabber is the place to go to for pricing hard drives en masse. I'm looking only at 7200 RPM drives here (faster)

Western Digital Caviar WD800BB 80GB $37 shipped - ATA-100 interface, make certain (from DELL) that this will work in your system (also ask about ATA-133). It's not important that you cannot run at full speed - all that's important is that these drives will work on your system. You may have ATA-33 or ATA-66. I *believe* that faster drives will work (at the slower speed).

Look at the "ultra ATA" listings at the left (if DELL has approved ATA-100 and ATA-133 for your system). Our 40 GB drive is available for $35 shipped, a Maxtor 120 GB 7200 is available for $40 shipped, and the 160 GB drive for $45 shipped. Clearly, you can keep going up - any of these drives should fit in your system, and you may want a 320 GB drive (though I don't know why). 250 GB looks like the most GB for the $ (Hitachi, $49 shipped, ATA-133, and 8 MB cache, which should make it even faster than the others). Avoid those "Magnetic Data" drives - this company is unknown to me. Make certain to get a 7200 RPM, and (again) make certain that ATA-100 and ATA-133 will work in your system.
** VIDEO **
Another problem is your video, which (I believe) uses shared RAM. Updating your video card should improve your game performance. You are restricted to a PCI bus (do *not* get anything that says PCIe or x16 or AGP). Again, pricegrabber is good for an overview

Why does nero 8 fail to burn my dvds while i use it with dvd shrink?
after dvd shrink finishes its bakup and it says to insert a blank disk, after about a few minutes at being at 0% burn it says burn fail and i get this messege

No it is not worth it. At least 2 GB of Ram, you want like 80-120+ Hard Drive space, you want to go with a Dual Core CPU, you want a decent graphics card, how much were you going to pay for that anyway? I say not worth it in any case.
